Section Staining for Electron Microscopy
Epon epoxy resin was introduced as an embedding medium for biological electron microscopy largely through the independent efforts of Finck (1) and Luft (2). The usual recipe involves three components: Epon 812 resin (Epikote 812, in England), an anhydride hardener, and an accelerator such as DMP-30. Since the combination of Epon with dodecenyl succinic anhydride (DDSA) was thought to produce a ...
متن کاملA New Epoxy Embedment for Electron Microscopy
A new epoxy embedding mixture has been developed utilizing Maraglas 655 and Cardolite NC-513 with benzyldimethylamine (BDMA) as a curing agent. This epoxy mixture permits cellular preservation comparable to that obtained with Epon 812, ease of preparation of tissues, a wide range of miscibility, low viscosity, and, most important, ease of sectioning on a Porter-Blum microtome. In contrast to Ep...
متن کاملA routine method for embedding animal tissues in Spurr resin for electron microscopy.
The epoxy resin epon 812 has been used in this department to embed a variety of normal and pathological tissues for a number of years. Unfortunately during the past 18 months sectioning qualities of polymerized epon blocks have become variable in an unpredictable manner. It is possible that the batch variation in the quality and composition of the dodecynl succinic anhydride (DDSA) could be res...

A new water-soluble embedding agent, tr ial p roduc t CIBA X 133/20971 has recently been in t roduced for selected electron microscopic investigations (1). D u r c u p a n subsequently proved to be miscible wi th the water-insoluble epoxy resin Araldi te 2 which is commonly used for tissue embedding (2). In this way, an imal tissues can be embedded in Araldi te wi thout being b rough t into con...
متن کاملEpoxy Resins in Electron Microscopy
A method of embedding biological specimens in araldite 502 (Ciba) has been developed for materials available in the United States. Araldite-embedded tissues are suitable for electron microscopy, but the cutting qualities of the resin necessitates more than routine attention during microtomy. The rather high viscosity of araldite 502 also seems to be an unnecessary handicap. The less viscous epo...
متن کاملImmunolabelling of prolactin at ultrastructural level using the protein A-gold technique on Epon-embedded tissue.
The use of the colloidal-gold technique in electron microscopy immunocytochemistry has provided important information on the in situ localisation of intracellular antigens. We have developed a post-embedding technique for prolactin localisation on resin-embedded human pituitary tissue sections by the use of the protein-A gold conjugate. Human pituitary tissue obtained at autopsy was processed f...
